The incident occurred in the northbound lanes of I-5 near South Hudson Street around 11:15 a.m.
One vehicle was on its side with a person trapped inside. Ambulances also transported two other victims with non-life-threatening injuries, officials said.
Three lanes of I-5 northbound were blocked at the scene, causing a three-mile backup. Two lanes have since reopened.
